【中文摘要】 對茶薪菇原生質體的分離及再生進行研究 ,結果表明以溶壁酶的去壁效果最好 ,不同酶組合可提高去壁效果 ,酶解液在 2 0 %時原生質體釋放量很高。酶解適宜溫度為 30℃ ,培養 4～ 5d的菌絲酶解 3h釋放的原生質體最多。以甘露醇作為滲透壓穩定劑 ,原生質體的釋放量及再生率均較高 ,再生率達到 5 %左右 ,經紫外線照射 2 0s,致死率即達 70 %以上
【英文摘要】 A study on protoplast isolation and regeneration from mycelia of Agrocybe chaxinggu showed that a superior system of protoplast preparation was determined by using 2.0% driselase as lytic enzymes at 25℃ for 3 hours,0.6 mol/L annitol as osmotic stabilizer could give higher protoplast yield and regeneration ratio,the protoplast regenerating ratio was about 3%～5%.With 20 seconds of UV-irradiation,the death rate of Agrocybe chaxinggu protoplast was up to 70%.
