【中文摘要】 從桃紅平菇單核菌絲分離出原生質體鋪于含0.6mol/l蔗糖的CM表面,經紫外線處理一定時間后,培養7—10天后長出的菌落被轉于MM之上,在其上生長十分緩慢或停止的菌落通過各種營養添加物測其營養需求,最后得到一個需要補充丙氨酸和天冬氨酸才能正常生長的雙缺突變體。試驗表明,該突變體能很好地分離出原生質體,且在含0.6mol/l蔗糖的MM上不能再生,因而具有較好的應用前景。
【英文摘要】 Protoplasts isolated from monokaryotic mycelia of Pleurotus salmonea-straminens were spread on CM containing 0.6 mol/l sucrose and irradiated by U. V. After 7—10 days, the regenerated colonies were transplanted on MM and those showing little growth were detected further with various kind of nutritional requirement., One auxotrophic mutant, double deficient in Ala and Asp, was obtained at last; from which a good protoplast yield could be obtained. It will be useful in protoplast fusion study because its protoplasts can not regenerate on MM containing 0.6 mol/l sucrose.
