【中文摘要】 應用RT PCR技術,研究了國產巴西蘑菇多糖(ABPS)對幾種細胞剌激因子(CSF)基因表達的影響。研究結果表明,人粒單祖細胞克隆剌激因子(GM CSF)和白細胞介素2(IL 2)擴增片段分別在255bp和500bp處出現明顯的條帶,而小鼠骨髓基質細胞表達的GM CSF則在369bp處出現一條帶。揭示國產巴西蘑菇多糖有誘導人臍血單個核細胞和小鼠骨髓基質細胞表達GM CSF和IL 2的作用,其濃度范圍為1～6μg/mL,最佳濃度為1μg/mL,從而證實了國產巴西蘑菇多糖促進粒單祖細胞生成和調節動物機體免疫功能的分子機理。
【英文摘要】 <Abstrcat> The effect of Chinese Agaricus blazei polysaccharide(ABPS)on the expression of cytokine genes was studied with reverse transcription-polymerase chain reaction technology. The results showed that the amplification segments of human granulocyte-macrophage colony stimulating factor(GM-CSF) and interlukin-2(IL-2) were 255bp and 500bp respectively ,while the GM-CSF from the mouse bone marrow stromal cells was 369bp. The experimental results suggested that ABPS could induce the expression of GM-CSF and IL-2 in human cord blood mononuclear cells and mouse bone marrow stromal cells,and the efficacious concentration of ABPS was 1～6μg/mL with the optimal concentration being 1μg/mL. The results also confirmed that ABPS could promote the colony forming unit-granulocyte-macrophage and regulate animal immune function.
