【摘要】 采用組織分離法,對東寧縣的松口蘑進行了分離培養。通過顯微觀察及RAPD-PCR技術對松口蘑子實體及其相應的培養物之間進行了親緣關系鑒定。結果表明,菌褶為松口蘑組織分離的最適宜部位,其最佳分離培養基為PWJ,即PDA+黑木耳二級種浸出液+麥麩。RAPD-PCR結果顯示,松口蘑子實體與分離物之間的DNA指紋相似系數在0.97～1.00之間,表明松口蘑子實體與其相應的培養物在種質上是一致的。 更多還原
【Abstract】 In this study,the mycelia of Tricholoma matsutake from Dongning were cultivated by the method of tissue separation and identified using RAPD-PCR.The results showed that lamellae were suitable for the tissue separation and well induced on PWJ medium.By RAPD-PCR analysis,DNA fingerprint similarity coefficient was 0.97～1.00,that implied the mycelia were indeed from their corresponding fruiting body. 更多還原
