靈芝菌絲原生質體形成與再生的研究

摘要：本文探討了靈芝(Ganoderma lucidum)菌絲原生質體的制備及再生的條件。實驗結果表明：采用MYG為靈芝菌絲培養基，2%真菌溶壁酶(Lywallzyme)作脫壁酶，0.6mol／L甘露醇作滲透壓穩定劑；對培養6d的靈芝菌絲在30℃下進行酶解，酶解時間4-6h，可產生約3.0×106個／mL(0.1g菌絲)原生質體，并在添加靈芝子實體浸出汁的雙層培養基上實現了靈芝菌絲原生質體的再生。

Formation and Regeneration of Protoplast From
the Mycelia of Ganoderma lucidum

Abstract:The conditions of protoplast formation and regeneration from the mycelia of Ganoderma lucidum were studied.Experimental results indicated that 3.0×106／mL protoplast of the mycelia of Ganoderma lucidum was obtained under optimal conditions,which are described as follows:Using 0.6mol／L mannitol as osmotic stabilizer;2% Lywallzyme as the enzyme digesting the cell wall;Using 6 days old MYG liquid cultures of the mycelia Ganoderma lucidum；at 30℃,enzyme digesting 4～6 hours.When incubated on two layer plates of agar,the protoplasts regenerated and formed new mycelia of Ganoderma lucidum.
