【摘要】 構建了香菇135菌株（Lentinula edodes）菌絲生物量與其DNA含量的標準曲線。通過一系列的篩選工作,確定了標準菌絲的培養條件及DNA提取系統,得到了三個不同培養批次香菇135菌絲體構建的高重現性的關聯標準曲線,它們均具有較高的相關系數（>0.99）,統計構建標準曲線的所有90個數據,測定的單點誤差范圍為0.05%13.74%（<14%）。所得數據表明在操作范圍內,菌絲生物量與DNA含量之間存在很好的線性關系,每mg香菇135標準培養菌絲（干重）對應的DNA含量為6.1μg。
【Abstract】 Standard curves relating DNA content and mycelial biomass were constructed for Lentinula edodes,strain 135.Adopting a set of standardized mycelium growth conditions and DNA extraction procedures,standard curves relating DNA concentration with mycelial biomass for three separate batches of L.edodes,strain 135,exibited a correlation coefficient >0.99.The error range for a total of 90 determinations(18 samples,five replicates of each sample)was between 0.05%-13.74%.Our data indicated that,under ...
